Interaktion mellan värmechockfaktor 2 och talin 1 : evaluering av cellinjer som modellsystem

Tiivistelmä
Proteiner är biomolekyler som interagerar för att bland annat ge cellen dess struktur och katalysera reaktioner. En potentiell proteininteraktion finns mellan fokaladhesionsproteinet talin 1 (TLN1) och transkriptionsfaktorn värmechockfaktor 2 (eng. heat shock factor 2, HSF2). HSF2 och TLN1 saknar kända överlappande funktioner, men de har motsatt effekt på migrations- och invasionspotentialerna i prostatacancercellinjer. I denna avhandling pro gradu användes de humana cellinjerna RWPE-1, PC-3, MCF-10A, MCF-7, U2OS och U2OS HSF2 KO, en variant av U2OS som saknar HSF2, samt muscellinjen MEF. Cellinjerna evaluerades som modellsystem för att karaktärisera den potentiella interaktionen mellan HSF2 och TLN1. Cellinjerna RWPE-1 och MCF-10A representerade normala celler och PC-3 och MFC-7 representerade cancercellinjer från prostata- respektive bröstepitel. Utav cellinjerna med tydlig komplexbildning av HSF2 och TLN1 kunde endast RWPE-1 och PC-3 transfekteras effektivt. En möjlig gränsyta i HSF2 för HSF2-TLN1-interaktionen utreddes genom att i PC-3-celler uttrycka en variant av HSF2, där den potentiella gränsytan muterats. Utredningen av gränsytan i HSF2 gav otydliga resultat, och direkt interaktion mellan HSF2 och TLN1 kunde varken bevisas eller motbevisas. Dock identifierades PC-3-cellinjen som ett möjligt modellsystem, plasmider framställdes och protokoll för transfektion och koimmunprecipitation optimerades för framtida undersökningar kring den potentiella interaktionen.

There is potentially a protein interaction between the focal adhesion protein talin 1 (TLN1) and the transcription factor heat shock factor 2 (HSF2). HSF2 and TLN1 lack known overlapping functions, but they have opposing effects on the migratory and invasive potential in prostate cancer cell lines. In this master’s thesis the human cell lines RWPE-1, PC-3, MCF-10A, MCF-7, U2OS and U2OS HSF2 KO, a variant of U2OS which lacks HSF2, and the murine cell line MEF were used. The cell lines were evaluated as model systems for characterization of the potential interaction between HSF2 and TLN1. The RWPE-1 and MCF-10A cell lines represented normal cells while PC‑3 and MCF-7 represented cancer cell lines from prostate and breast epithelium, respectively. Out of the cell lines with clear complex formation of HSF2 and TLN1, only RWPE-1 and PC-3 could be effectively transfected. One possible interface in HSF2 for the HSF2-TLN1-interaction was investigated in PC-3 cells by expressing a variant of HSF2 in them where the interface had been mutated. The results from the investigation of the interface were inconclusive, and a direct interaction between HSF2 and TLN1 could neither be proven or disproven. Nevertheless, the PC-3 cell line was identified as a usable model system, plasmids were generated and protocols for transfection and co-immunoprecipitation were optimized for future studies regarding the potential interaction.
